After qualifying for the M.D. degree at Basel in 1855, His became an instructor in histology before his appointment in 1857 as professor of anatomy and physiology at the university. In 1872 he moved to Leipzig to became professor of anatomy and head of the Institute of Anatomy. This work alone would have established His as one of the top ranking histologists and embryologists of the nineteenth century. Concerning the embryology of the chick, the book contains illustrations drawn by His and mentions techniques that he was among the first to employ. His' methodology included the use of paraffin as an embedding mass for embryological purposes and the use of a microtome that he developed for sectioning specimens.
