Winkler studied medicine at Utrecht, graduating in 1879. He spent the early part of his career at Utrecht where he practiced, performed research, and taught at the university. In 1896 he accepted an appointment at Amsterdam and nineteen years later returned to Utrecht, where he spent the remainder of his career. Winkler was a prolific author and published many papers on neurology, psychiatry, criminal anthropology, and pathology of the brain. One of the most important was this paper on the central pathways of the eighth cranial nerve which also appeared in Verhandelingen der Koninklijke Akademie van Wetenschappen te Amsterdam.
